indications

Ketosan drug is indicated in the following cases:

  • Preventing bronchial asthma (asthma, asthma, asthma in the case of seasonal fever and allergic bronchitis). At the beginning of treatment should continue to use basic asthma medications: bronchodilator, theophyllin or corticosteroids.
  • Treatment of allergic rhinitis, systemic allergies, urticaria, atopic dermatitis, rashes, allergic conjunctivitis.
  • Note: Ketotifen is not indicated to treat acute asthma attacks.

    Pharmacokology

Ketotifen is a non -compatible histamine inhibitor (H1 - receptor) inhibitors, relatively selective and a mast cell protective agent. Ketotifen is an oral medication that has the main effect on the respiratory system, is a medication for asthma without bronchodilator and has anti -allergic effect.

Inhibit the release of chemical mediators that cause allergens from mast cells and related cells in allergic reactions such as histamine, leukotrien C4 and 04 (SRS - A) and PAF.

H1 - histamine receptor inhibitors and phosphodiesterase inhibitors and the results increase the content of the Camp in Mast cells.

Preventing the progression of respiratory allergies, inhibiting PAF platelet activation (platelet activation) or due to nerve stimulation after using sympathetic stimulants; Preventing the gathering of eosinophil and platelets due to the respiratory tract, inhibiting the increase in the eosinophil, thus inhibiting the eosinophil line into the inflammatory place.

Long -term treatment effect on mild allergy bronchial asthma has been shown in clinical trials. Ketotifen continuous use every day helps reduce frequency, severity and prolonged symptoms of asthma and asthma attacks, helping to reduce the daily needs of coordinated asthma medications (theophylllin, β2 ...) without reducing lung function (FEV1, FVC and PEFR).

Ketotifen does not work to treat acute asthma attacks.

    Pharmacokinetics

    absorption

Ketotifen is well absorbed through the gastrointestinal tract (≥ 60%) after drinking. But bioavailability is only about 50% due to the first metabolism of the liver. The peak concentration in plasma is achieved after 2-4 hours, the absorption time is 1 hour. Food does not affect the bioavailability of the drug.

    Distribution

The ratio of cohesion to plasma protein is 75%.

    Metabolism

Ketotifen experienced the first liver effect (approximately 50%). Main metabolites in plasma and urine in the form of ketotifen - n - glucuronid. The metabolism of the drug in children is similar to adults but the clearance is higher. Therefore, in children> 3 years old, daily dose is similar to adults. When it is necessary to use drugs for children

    Elimination

Ketotifen is eliminated through 2 sections:

    Semi -cancellation time T1/2:

  • Distribution phase: 3 - 5 hours.
  • Erimization phase: 21 hours.

Ketotifen excreted mainly through urinary tract: 1% of the original form and 60-70% of the metabolic form is not active within 48 hours. The waste time is about 21 hours.
