Pharmacokology
Loratadin is a 3 -round antihistamine that has a long -lasting effect with selective antagonistic activity on H1 receptor.
pharmacokinetics
Loratadin is quickly absorbed through the gastrointestinal tract after drinking, the peak concentration in plasma is about 1 hour.
Increased bioavailability and time to reach peak plasma concentrations slowly when shared with food. Loratadin is metabolized. The main metabolites are desloratadine with effective antihistamine activity.
The average selling time was reported by loratadin and desloratadin, respectively 8.4 and 28 hours. Loratadine binds about 98% of plasma proteins; Desloratadine binds less. Loratadine and its metabolites are detected in breast milk, but it does not pass the brain barrier in significant amounts. Most of the drug dose is excreted in urine and feces in the same amount, mainly in the form of metabolites.
Loratadin's distribution does not see significant changes in patients with severe renal impairment and hemorrhage is not a measure of effectively eliminating loratadine or diameting desloratadin from the body.

Handling
Treatment of ordinary Loratadin overdose includes symptomatic treatment and supportive treatment, starting immediately and maintaining as needed.
In case of acute loratadin overdose, empty stomach should be immediately using ipeca syrup. Taking activated carbon after vomiting can effectively prevent Loratadin's absorption. If vomiting is ineffective or contraindicated (such as with a coma, convulsions), it is possible to conduct a stomach rinse with a 0.9% Naci solution if there is an intravotic tracheal tube to prevent the inhalation of substances in the stomach. Saline detergents have the effect of quickly diluting the substances contained in the intestine.
What to do when you forget the dose? However, if close to the next dose, skip the forgotten dose and take the next dose at the time as planned. Do not drink twice as prescribed.

Drug interaction
simultaneous treatment of loratadine and cimetidine leads to increased loratadine concentration in plasma 60%, due to cimetidine metabolic inhibits Loratadine. This has no clinical manifestations.
Simultaneous treatment of loratadine and ketoconazole leads to an increase in loratadine levels in plasma 3 times, due to CYP3A4 inhibition. This has no clinical manifestations because Loratadin has a broad treatment index.
Concomitance of Loratadin and erythromycin leads to increased plasma Loratadin levels. Loratadin's AUC increased by 40% and the AUC of Descarboethoxyloratadine increased by 46% average compared to only Loratadine treatment. On the center of the center, there is no change in the QTC range. Clinically, there is no manifestation of loratadin's safety changes, and there is no notice of sedative effects or fainting phenomena when treating with this drug.
