Pharmacokinetics
absorption
Acetaminophen is absorbed quickly and completely after drinking. Peak concentration in plasma reaches about 15 minutes to 2 hours after drinking. Acetaminophen's oral use is about 80% and does not depend on the dose of about 5 to 20 mg/kg.
Distribution
Acetaminophen is almost not linked to plasma proteins. The concentration of acetaminophen in saliva is equivalent to plasma. The total concentration in the blood is 20% higher and 20% lower in breast milk than plasma concentrations. Acetaminophen passes the placenta.
Metabolism and elimination
Acetaminophen is metabolized mainly in the liver and the total clearance of the body is about 5 ml/kg/minute. The clearance of acetaminophen decreases and the selling time increases after overdose of the liver toxicity.
Prolonged half -life for 4 hours often shows the risk of liver damage.
Two to five percent of acetaminophen treatment dose is excreted in a constant form in urine.
